I have no idea what is going on in your system. But I would suggest that you should install a rt kernel (5.10.0-16-rt-amd64 #1 SMP PREEMPT_RT Debian - I am using this one), so you can run in a very low latency and direct from alsa (which is better). I am running PT at 128 polyphony and at 128 samples - 2.9 ms without any glitch, I could could go further but I prefer to keep safe.
You can try and I hope that works.
