YES! "Downgrade" USB 3 to USB 2 via a hub solves the problem.
Thanks for your input. So glad I kept that old USB hub.....
And I will relate the story to the Focusrite-Engineers. It is about time they realize there are a few W10 users, and that USB has evolved to 3.0.

I had the same goal and the same problem with the same configuration.
It seems that Focusrite USB 2.0 Audio Driver won't work correctly with USB 3.0 port on NUC under Windows 10.

Solution that I've found ist to insert a kind of "converter" between NUC and Focusrite - I had an old Logitech USB 2.0 hub.
Although the hub is active, I don't use external power suply for it. Since than I didn't experience any problem with autostart.

My hardware:
INTEL NUC with Windows 10, MIDI-keyboard connected via USB, DA-converter Forte.
Software: ASIO as well as Focusrite USB 2.0 Audio Driver.

I start Pianoteq in autostart-mode with the following settings:
Audio-Device Type: ASIO
Output: Focusrite USB 2.0 Audio Driver

Pianoteq will complain it could not create I/O-Buffers. So I need to first go back to Windows Audio, and then again change for ASIO.

This is annoying, because I want the software to start unattended when I switch on my computer.

Any idea is welcome.
